Introduction *
Gliomas constitute a heterogeneous group of primary brain tumors with variable prognosis. This study used data from the cBioPortal for Cancer Genomics aiming to describe the demographic, clinical, and molecular characteristics of patients with glioma and to evaluate their impact on survival outcomes.
Material & Method *
Data of 530 patients were analyzed for the following variables: demographic characteristics, histologic subtypes, molecular markers (including IDH1 mutations and tumor mutational burden), initial symptoms, and survival outcomes. We assessed the impact of clinical and molecular factors on overall survival (OS) and disease-free survival (DFS) using descriptive statistics, Kaplan-Meier survival analysis, and Cox regression models.
Results *
55.0% of the patients were males with a mean age of 42.8 years at diagnosis. The most frequent subtypes were astrocytoma (37.2%), oligoastrocytoma (25.3%), and oligodendroglioma (37.4%). 72.8% of patients had IDH1 mutation and most frequently in oligodendrogliomas (83.8%). The mean OS was 32.4, ranging from 36.9 months in oligodendrogliomas to 29.3 months in astrocytomas. Sensory symptoms at presentation showed the longest median survival (154.2 months), while the shortest (44.4 months) seen with mental status changes. Histologic subtype, IDH1 mutation, and initial symptoms were identified as significant predictors of survival using multivariate analysis.
Conclusion *
This study highlights the influence of molecular profiling, histologic subtype, and initial symptoms on survival outcomes in glioma patients. IDH1 mutations and oligodendroglioma histology were associated with improved survival, while mental status changes were implicated in poor outcomes. Individualized management based on molecular, histologic and clinical characteristics is needed to optimize care and improve survival outcomes.
