Introduction *
Magnolia salicifolia (MS) is a tree native to Japan, and its essential oil is popular as a Japanese aroma oil. In this study, we investigated the antitumor effects of MS essential oil against breast cancer cells.
Material & Method *
Breast cancer cells were cultured in a 96-well plate, and essential oil was added dropwise only to the central well. By incubating the plate at 37 degrees for 24 hours, the effect of the volatile components of the essential oil on the surrounding breast cancer cell growth ability was examined using the MTT assay and fluorescence microscope.
Results *
Forty kinds of essential oils were tested, and the essential oils of MS was found to have strong antitumor effects against mouse breast cancer cells (4T1). The antitumor effect presents in the evaporated components of MS essential oil decreased in a distance-dependent manner. MS contains a large amount of low molecular weight compounds such as 1,8-Cineole, Neral, Geranial, and trans-Anethole. The anti-tumor effect of Citral (Neral + Geranial), which is the most abundant of these compounds, was 8.9 times that of 1,8-Cineole, and was the most important antitumor factor in MS essential oil. It was shown that the mechanism by which Citral induces cancer cell death is through the release of cytochrome C and activation of caspases 3 and 9 following a decrease in mitochondrial membrane potential induced by Bax.
Conclusion *
The transpiration components of MS is thought to induce inhibition of breast cancer cell proliferation by Citral.
